Abstract. Mucin-type O-linked glycosylation is a fundamental post-translational modification that is involved in a variety of important biological processes. However, the lack of chemical tools to study mucin-type O-linked glycosylation has hindered our molecular understanding of O-linked glycans in many biological contexts. As noted above, the defining feature of mucin O-linked glycosylation compared to other types of glycosylation is the initial addition of GalNAc to Ser/Thr residues of protein substrates. 37 One useful group of metabolic inhibitors of mucin O-linked glycosylation utilizes GalNAc analogs as decoy primers that suppress elaboration of O-linked glycans. 50,51 These unnatural sugars exploit two 2005-11-01 Plasma membrane signaling mucin that promotes activation of the MAPK for the filamentous growth pathway.
